I think it's "eaker/eeker" from Summer School.

Melissa McCarthy's husband is Ben Falcone. The actor in the Target commercial sure looks like a lot like him. I couln't find anything online that credits him in the ad, though. When I first watched the commercial I thought it was the guy who played Neil Kellermann in Dirty Dancing (it wasn't).
