While it may be natural to think locational AC is better, I am debating if it would be constantly fighting with neighboring rooms where AC is not running, as opposed to Central AC, which can equalize the whole house.

I think the first 2 answers you got are based on guesses. I don't know how to figure this out. Have you call your local electric company. They probably have the answer & please come back & let us all know.
My central A/c broke & I paid $300 to have it repaired & it still didn't work well. I live in a 2400 sq. foot house & I am usually home alone each day. When I was younger I wanted to clean the whole house & be free to go anywhere in the house & now I stay in one place most of the time. So I use window unity. My electric bills seems low but it could also be that I have not had to run it as much so I am not sure how to give you an accurate answer.
I do think the electric co could tell you how much it cost to run each unit but it would not be able to tell how that works with you lifestyle & your house.
I sit in my living room & the ac in the dinning room keeps me cool. My downstairs is open but the cool air stays up here.
If you keep the door closed on your single room, no problem.
